Gladwell Digs into Cancer Drug Research Difficulties

Remember this statistic when you’re reading an enthusiastic drug company press release: 90% of the drugs studied in humans to fight cancer (and other diseases) show no difference between the control and treatment arms.

That’s not a new stat, nor is the notion that cancer research is a heartbreaker of a field. But Malcolm Gladwell takes a characteristically deep dive (subscription required) into the problem in this week’s New Yorker, pointing out that given the pitfalls of cancer drug development, a “master code behind the disease” may not be waiting to be cracked. “Perhaps there is only what can be uncovered, one step at a time, through trial and error,” he writes.

Gladwell uses the story of elesclomol, a compound developed by Synta Pharmaceuticals, to make his point: the drug, which causes cancer cells so much oxidative stress that they die, showed promise in the lab. It eventually became the subject of a phase 3 melanoma trial funded by GlaxoSmithKline. But in February of last year, the trial was halted over safety concerns. (The company recently announced that the FDA has allowed it to resume clinical development of elesclomol.)

Despite the cautionary tale, Gladwell seems to come down on the side of the mass screening approach – randomly combing over drugs to see if they might be suitable against a range of diseases – that led to elesclomol’s development. The alternative, rational design, is epitomized by the successful development of the drug Herceptin: figuring out the problem to be solved, in that case the overproduction of the Her-2 protein by certain breast cancers, then designing a drug to address the problem.

But, as Gladwell writes, “working backward to a solution requires a precise understanding of the problem, and cancer remains so mysterious and complex that in most cases scientists don’t have that precise understanding. Or they think they do, and then, after they turn off one mechanism, they discover that the tumor has other deadly tricks in reserve.”

Speaking at an international water conference, the CEO of the Gates Foundation called for water efficiency while improving agriculture yields. Jeff Raikes of the Gates Foundation talked about the science breakthroughs in modified seeds that produce more food with less water.

Ralkes also showed slides of dried up lakes and rivers in Asia and Africa and how this will cause conflicts in the future.

From KOMO news, AP writer Eric Olson recieved a bunch of quotes from the Gates Foundation CEO.

“I see the linkage of the water crisis and the future of global poverty,” he said, “yet I don’t see the general awareness of this issue. I don’t hear the talk of securing water for food.

“We worry about terrorism, yet do we understand that the battles over resources are the most devastating?”

Raikes echoed the notion that many of the 20th century’s conflicts were based on oil, and many of this century’s conflicts will be over water.

Without better farming practices and water management in impoverished areas, the amount of water required to meet global demand for food will almost double by 2050, Raikes said.

“We have to get more food on the same land with the same or less amount of water,” Raikes said.

The world population is projected to grow from 6 billion to 9 billion over the next 40 years, with the biggest increase in developing nations. Improving diets in developing nations such as India and China will further increase food demand, he said.

A dedicated hosting service, dedicated server, or managed hosting service is a type of Internet hosting in which the client leases an entire server not shared with anyone. This is more flexible than shared hosting, as organizations have full control over the server(s), including choice of operating system, hardware, etc. Server administration can usually be provided by the hosting company as an add-on service. In some cases a dedicated server can offer less overhead and a larger return on investment. Dedicated servers are most often housed in data centers, similar to colocation facilities, providing redundant power sources and HVAC systems. In contrast to colocation, the server hardware is owned by the provider and in some cases they will provide support for your operating system or applications.

Bandwidth refers to the data transfer rate or the amount of data that can be carried from one point to another in a given time period (usually a second) and is often represented in bits (of data) per second (bit/s). For example, visitors to your server, web site, or applications utilize bandwidth as the traffic moves from your server to the Internet and vice versa. Connectivity refers to the “access providers” that supply bandwidth, or data transfer rate, through various connection points across a network or footprint to one or multiple data centers where dedicated servers are housed.

Bandwidth measurements are defined (per telecom standards) as the following:

* First – 95th (measured using average bits and speed of transfer)
* Second – Unmetered (measured in speed or bits)
* Third – Total Transfer (measured in bytes transferred)

95th Method: line speed, billed on the 95th percentile, average or peak usage, refers to the speed in which data flows from the server or device. Line speed is measured in bits per second (or kilobits per second, megabits per second or gigabits per second).

Unmetered Method: The second bandwidth measurement is unmetered service where providers cap or control the “top line” speed for a server. Top line speed in unmetered bandwidth is the total Mbit/s allocated to the server and configured on the switch level. For example, if you purchase 10 Mbit/s unmetered bandwidth, the top line speed would be 10 Mbit/s. 10 Mbit/s would result in the provider controlling the speed transfers take place while providing the ability for the dedicated server owner to not be charged with bandwidth overages. Unmetered bandwidth services usually incur an additional charge.

Total Transfer Method: Some providers will calculate the Total Transfer, the measurement of actual data leaving and arriving, measured in bytes. Measurement between providers varies, though it is either the total traffic in, the total traffic out, whichever is the greater or the sum of the two.

One of the reasons for choosing to outsource dedicated servers is the availability of high powered networks from multiple providers. As dedicated server providers utilize massive amounts of bandwidth, they are able to secure lower volume based pricing to include a multi-provider blend of bandwidth. To achieve the same type of network without a multi-provider blend of bandwidth, a large investment in core routers, long term contracts, and expensive monthly bills would need to be in place. The expenses needed to develop a network without a multi-provider blend of bandwidth does not make sense economically for hosting providers.

Many dedicated server providers include a service level agreement based on network uptime. Some dedicated server hosting providers offer a 100% uptime guarantee on their network. By securing multiple vendors for connectivity and using redundant hardware, providers are able to guarantee higher uptimes; usually between 99-100% uptime if they are a higher quality provider. One aspect of higher quality providers is they are most likely to be multi-homed across multiple quality uplink providers, which in turn, provides significant redundancy in the event one goes down in addition to potentially improved routes to destinations.

Bandwidth consumption over the last several years has shifted from a per megabit usage model to a per gigabyte usage model. Bandwidth was traditionally measured in line speed access that included the ability to purchase needed megabits at a given monthly cost. As the shared hosting model developed, the trend towards gigabyte or total bytes transferred, replaced the megabit line speed model so dedicated server providers started offering per gigabyte.

Prominent players in the dedicated server market offer large amounts of bandwidth ranging from 500 gigabytes to 3000 gigabytes using the “overselling” model. It is not uncommon for major players to provide dedicated servers with 1Terabyte (TB) of bandwidth or higher. Usage models based on the byte level measurement usually include a given amount of bandwidth with each server and a price per gigabyte after a certain threshold has been reached. Expect to pay additional fees for bandwidth overage usage. For example, if a dedicated server has been given 3000 gigabytes of bandwidth per month and the customer uses 5000 gigabytes of bandwidth within the billing period, the additional 2000 gigabytes of bandwidth will be invoiced as bandwidth overage. Each provider has a different model for billing. As of yet, no industry standards have been set.

What’s the difference between recordable DVD disc formats?

There are seven formats of DVD recordable disc available. They offer different features and not all are compatible with all recorders. We’ll steer you through the maze and help you select the right disc for you.

Standard single-layer recordable DVD discs can store an hour of footage at their highest quality setting. Picture and sound quality are still very good if you extend this to two hours.

You can push them further to record three, four or even eight hours of footage on a disc but picture quality becomes increasingly poor. Dual layer discs have double the recording capacity of single layer discs.

DVD+R and DVD-R

These discs are cheap, write-once-only discs which you cannot re-record on. However, new recordings can be added later to the remaining blank section. Recordings cannot be erased though you can hide them by deleting their titles from the contents listing that’s shown when you first play a disc.

To play the disc in machines other than the one you recorded with, the disc has to be ‘finalised’ meaning that no further recordings can be made to that disc. Dual-layer versions of these discs have twice the recording capacity – most modern DVD recorders can use them.
DVD+RW and DVD-RW

These are re-recordable versions of DVD-R and DVD+R discs – you can overwrite existing recordings with new ones.

DVD-RW has two different recording modes.

In ‘video’ mode, recordings are made in order you make them, with new ones added to the remaining blank section. Recordings can be deleted but if you re-record in the blank space you must ensure the recording will fit or it’ll copy over the next recording.

In ‘VR mode’ it avoids this by jumping over subsequent recordings to the next available space. You can also start watching a programme before it’s finished recording and it also allows more editing features to be used, such as joining or cropping recordings. However, not as many DVD players are compatible with discs recorded in VR mode.

On DVD-Ram discs, programmes aren’t saved in the order you record them but any available blank space is used. Recording may start off in one part of the disc and, when that’s full, it will skip to the next available blank space rather than needing a single space on the disc big enough to store the whole of the recording.

They offer various title-erase and edit facilities and are the most versatile type of disc in this respect. You can also start watching a programme before it’s finished recording. They sometimes come in a protective caddy to prevent damage to the disc surface, which is more delicate than other types of disc.

DVD-RAM disc compatibility is less-common than with the other formats.
